A team from the SPI (DMTS) investigated the effects of hypoxia on the interface between the choroid plexus and cerebrospinal fluid using organoids derived from human pluripotent stem cells. The aim was to investigate how cerebral hypoxia at birth, which causes hypoxic-ischaemic encephalopathy, disrupts this cerebral barrier, which has not yet been extensively studied.

Understanding protein-nanoplastic interactions – a step toward combating pollution?

Peptides and proteins adsorb onto plastic nanoparticles, forming a corona. Researchers from I2BC and CEA-Iramis, using molecular modeling, reveal distinct adsorption behaviors depending on their amino acid sequences. They thus establish a mechanical basis for predicting peptide-plastic interactions, a crucial element for assessing the risks these interactions pose to health and ecosystems.

An I2BC team has revealed the structure of the C-terminal transmembrane region of the SARS-CoV-2 protein nsp3, essential for virus replication. This region self-assembles into hexamers that form molecular pores on the surface of double-membrane vesicles, the sites of viral genome replication. Nsp3 therefore appears to be a promising therapeutic target.

DNA repair machinery more flexible thanks to PAXX?

An international collaboration led by researchers from I2BC/CEA-Joliot and IPBS (Toulouse) has solved the atomic structure of molecular assemblies involved in the repair of DNA double-strand breaks. The diversity of assemblies involving the PAXX protein identified in this study highlights the great flexibility of the repair machinery, which could reflect its ability to adapt to biological conditions. The results were published in Science Advances.

A study led by a team from the Institut Joliot (I2BC/B3S) reveals an unusual mode of interaction between a viral deubiquitinase and a ubiquitin, which appears to be optimized to finely control virus replication.

Composition of the microbiota of animals in natura: a challenge met by mass spectrometry

A team from SPI in Marcoule, in collaboration with INRAE, characterized the intestinal microbiota of a small crustacean, Gammarus fossarum, used as a sentinel of water quality in France, by combining proteogenomics and metaproteomics approaches, thus opening up interesting perspectives for the analysis of host/microbiota interactions in other animal models.

Lamins and Progeria: What Does Structural Biology Tell Us?

A team from the I2BC publishes in FEBS Journal an exhaustive review of the various mutations affecting the structure of type A lamins, proteins that make up the envelope of the cell nucleus (nuclear lamina), which lead to premature aging in people with the disease (Progeria).

SARS-CoV-2 detection: mass spectrometry shows its potential for rapidity.

A team from SPI in Marcoule detected by mass spectrometry, in 3 minutes and without specific reagents, signature peptides of the SARS-CoV-2 virus in clinical samples (nasopharyngeal swabs). It thus provides a proof of concept of the use of this method as a possible alternative to PCR, currently the reference method.
