Regulation of Fe–S cluster biosynthesis, whose failure causes a rare neurological disease, finally elucidated !

A team from I2BC has uncovered the key steps governing the assembly and regulation of iron–sulfur cluster biosynthesis, essential metallic cofactors involved in numerous biological functions. Defects in this process lead to pathologies such as Friedreich’s ataxia. These studies represent major advances toward the development of future therapies.

An international collaboration, led by the Institut du Cerveau et de la Moelle épinière – ICM (Brain & Spine Institute) and involving researchers from NeuroSpin, offers a new electroencephalogram (EEG) tool that helps the diagnosis of disorders of consciousness. Unlike other tools developed so far and dedicated to a few expert centers, “DoC-Forest” would be accessible to most of the hospitals worldwide.

What is the relationship between variants of the BRCA1 gene and susceptibility to cancer?

Research Teams from the Curie Institute, the Gustave Roussy Institute, as well as the biophysics teams from the Institute of Chemistry of Natural Substances and SB2SM (I2BC@Saclay) met to systematically identify structural defects and BRCA1 proteins encoded by variants of the BRCA1 gene, a gene predisposing to breast and ovarian cancer. Their results make it possible to propose a classification of these variants (neutral versus causal).

Protein modification: myristoylation finally decoded

An I2BC Research Team, in collaboration with the ICSN and the Ecole Polytechnique, has mapped for the first time, in man and the model plant Arabidopsis thaliana, the myristoylome, namely the whole proteins modified with a fatty acid, myristate. This modification is important because it targets these proteins to the membranes and thus contributes to their subcellular compartmentalization. These data represent an important resource, available to the community of biologists.

Collaborative work between the SBIGeM (I2BC@Saclay) and the Marseille Cancer Research Center, published in Molecular Cell, illuminates the mechanistic basis and genetic control of homologous recombination, a mechanism for repairing DNA breaks responsible for chromosome exchange during meiosis. However, canonical models do not explain the complexity of meiotic recombination profiles and this study, which reconstructs reaction intermediates, constitutes a major advance in the understanding of this mechanism.

Epigenetics: how to reach inaccessible genes?

Biologists have brought to light the operation of "chromatin remodellers", key enzymes in cells. They have discovered how the genetic material, condensed in the nucleus of the cell, needs to be remodelled so that the cell machinery can access the genes. This work, initiated by IBITECS/SBIGeM, coordinated by researchers from a CEA/CNRS/Université Paris-Sud laboratory, under an international collaborative project with Pennsylvania State University (USA) and Guangzhou University (China), was published in Nature on 4 February 2016.
